Every cell in your body lives inside a material: the extracellular matrix. In this full animated lecture we build the topic from the basics up to Master level, slowly and with repetition, so that it really sticks.
Part 1, the ECM itself: the four classes of components (collagens, proteoglycans, glycoproteins, elastin), the collagen triple helix and the Gly-X-Y rule, fibril assembly with quarter-staggered packing and 67 nm D-banding, collagen synthesis from gene to fibre, the proteoglycan cushion and osmotic swelling pressure, fibronectin and the RGD sequence, laminin and the blood-brain barrier, elastin, ECM functions, evolution, exon shuffling and the matrisome.
Part 2, cell-ECM contact: basement membranes versus interstitial matrix, cartilage and bone, ECM remodelling in bone and angiogenesis, matrix metalloproteinases and the cysteine switch, integrins and the integrin family wheel (24 integrins), integrin activation (bent, extended, clustered), inside-out and outside-in signalling, the adhesome, nascent, focal and fibrillar adhesions, cell motility on aligned matrix, cell polarity, epithelial-mesenchymal transition, anchorage dependence of growth and anoikis, and mechanotransduction.
Part 3, ECM diseases: ECM homeostasis (arthritis versus fibrosis), osteogenesis imperfecta, epidermolysis bullosa, scurvy, how ECM gene mutations act (haploinsufficiency, ER stress, dominant negative effects), and the ECM in cancer with tenascin as a therapeutic target.
Every term is defined when it first appears, and every process is shown in motion: the triple helix twisting, fibrils packing, integrins unbending, blood vessels sprouting, bone being tunnelled and refilled.
