The fluorescent labeling of proteins has tremendous value for in vitro biophysical experiments as well as for cell-based imaging. In all of these experiments, incorporation of intrinsically fluorescent non-canonical amino acids via GCE can provide significant advantages over labeling using fluorescent protein fusions or chemical modification. I will discuss these advantages as I describe the incorporation of acridonylalanine (Acd) through GCE in bacteria for biophysical analysis of purified proteins and in mammalian cells for imaging. A variety of applications will be described, using Acd alone and in combination with FRET partners.
