Tips
The MS Chi-Cheemaun ferry takes you from Tobermory to Manitoulin Island. It's about 2 hours each way and runs a couple times a day.
Highway 6 is pretty calm on Manitoulin Island, but past Little Current it gets very busy until Espanola. The shoulders are generally good but beware sandy spots and tight turns. This is all technically part of the Great Lakes Waterfront Trail, but be careful.
Manitoulin Eco Park has really cheap campsites (about $20/night) and includes running showers and washrooms. There is amazing stargazing especially in its DarkSky preserve. Sometimes the staff will host stargazing and telescope events. Highly recommended.
There are a couple more campsites on the island, but they can be more expensive and/or require a minimum of 2 nights booking.
There are generally very few stores on this route. Sheguiandah, Little Current and Espanola are the major supply points.
I highly recommend splitting this route and staying overnight at Espanola.
From Espanola to Sudbury, you will need to go on Highway 17, but only for 7 km. Otherwise there are some very good sideroads you can stay on as you approach Sudbury.
As you approach Sudbury, you will be forced to stay on Highway 17 for longer stretches. It can be scary doing this in the dark. You can try to cycle through the surrounding the suburbs, but inevitably you'll be funneled back onto Highway 17. Avoid doing all this in the dark.
Sudbury itself is pretty bicycle friendly. The Northland Express bus terminal is on the east side of the city, so you'll need to cycle across. It is relatively easy to do so.
You can take the Northland Express back to the GTA. The terminal can distribute bike boxes and tape, you'll need to dismantle your bike a bit to do so. Make sure to have your bike packed up before the bus departs.
I suggest booking your ticket in advance. Check Northland Express's current bike policy. I had to call them to book my bus ticket with bike box (their website doesn't include the bike box option). The bike box costs extra. You can attempt to save that cost by sourcing a bike box from a nearby bike store, but IMO it's not worth the hassle.
