The basic thawing procedure is similar to that used for conventional cell lines. However, recovering a CRISPR library requires one additional consideration: maintaining sufficient sgRNA coverage.
In this video, we demonstrate the recovery of hGeCKO Library A#2 in THP-1 and walk through the key steps, including:
• Calculating the required cell number based on sgRNA coverage
• Processing multiple cryovials as a group
• Minimizing DMSO exposure during thawing
• Pooling and evenly distributing the recovered cells
• Evaluating cell morphology 24 hours post-thaw
For a CRISPR library, the goal is not simply to recover viable cells, but to recover enough cells to maintain adequate library representation.
