Structural Outline of the Glycolytic Pathway (Glycolysis) Glycolysis is a metabolic pathway in which one molecule of glucose is broken down into two molecules of pyruvate through a sequence of ten enzyme-catalysed reactions. It occurs in the cytoplasm of cells and provides energy in the form of ATP and NADH. Glycolysis can operate whether oxygen is available or not, although the subsequent fate of pyruvate differs. The 10 Steps of Glycolysis Phase 1: Energy Investment Phase 1. Glucose → Glucose-6-phosphate Enzyme: Hexokinase or glucokinase Uses 1 ATP. 2. Glucose-6-phosphate → Fructose-6-phosphate Enzyme: Phosphoglucose isomerase. 3. Fructose-6-phosphate → Fructose-1,6-bisphosphate Enzyme: Phosphofructokinase-1 (PFK-1) Uses 1 ATP and is a major regulatory step. 4. Fructose-1,6-bisphosphate → Glyceraldehyde-3-phosphate (G3P) + Dihydroxyacetone phosphate (DHAP) Enzyme: Aldolase. 5. DHAP → Glyceraldehyde-3-phosphate Enzyme: Triose phosphate isomerase. At this stage, two molecules of G3P proceed through the payoff phase per glucose molecule. Phase 2: Energy Payoff Phase 6. G3P → 1,3-Bisphosphoglycerate Enzyme: Glyceraldehyde-3-phosphate dehydrogenase. Produces NADH; two NADH are formed per glucose. 7. 1,3-Bisphosphoglycerate → 3-Phosphoglycerate Enzyme: Phosphoglycerate kinase. Produces ATP; two ATP are formed per glucose. 8. 3-Phosphoglycerate → 2-Phosphoglycerate Enzyme: Phosphoglycerate mutase. 9. 2-Phosphoglycerate → Phosphoenolpyruvate (PEP) Enzyme: Enolase. Releases water. 10. Phosphoenolpyruvate → Pyruvate Enzyme: Pyruvate kinase. Produces ATP; two ATP are formed per glucose. Overall Energy Yield per Glucose • ATP consumed: 2 • ATP produced: 4 • Net ATP: 2 • NADH produced: 2 • Final product: 2 pyruvate molecules Under anaerobic conditions in human cells, pyruvate may be converted into lactate to regenerate NAD+. Under aerobic conditions, pyruvate can enter mitochondria and be converted into acetyl-CoA for further oxidation. Key Regulatory Enzymes The three major irreversible regulatory steps are catalysed by hexokinase or glucokinase, phosphofructokinase-1, and pyruvate kinase. PFK-1 is a key control point influenced by cellular energy status and regulatory metabolites. Conclusion Glycolysis consists of ten enzyme-catalysed steps divided into energy investment and energy payoff phases. It is essential for ATP production, glucose metabolism, and the provision of intermediates for other biochemical pathways. #Glycolysis #GlycolyticPathway #Biochemistry #GlucoseMetabolism #ATP #NADH #CellularRespiration #MedicalEducation
