Disruption of sphingolipid homeostasis and signaling has been implicated in many conditions including cardiovascular diseases, but mechanisms governing cellular sensing of SL and subsequent regulation of their biosynthesis remain unknown. Here, Linda Sasset of Weill Cornell Medicine, USA discusses interactions between S1P-S1PR signaling and ORMDLs, key regulators of the first step of sphingolipid synthesis. She further discusses how disruption of this axis results in endothelial dysfunction, which is an early event in the onset of cardio- and cerebrovascular diseases.
