Title: Revitalizing Mitochondria against Alzheimer’s disease: The Synergistic Effects of a Nutraceutical Combination with urolithin A
Abstract:
Urolithin A (UA), a gut microbiota-derived metabolite of ellagitannins, has emerged as a potent nutraceutical compound with the ability to induce mitophagy, a selective autophagic process crucial for mitochondrial quality control. Our studies aimed at investigating the effects of UA on mitochondrial dynamics and cellular health, highlighting its potential in combating Alzheimer’s disease (AD). The studies conducted in the neuroblastoma cell line, BE(2)-M17, revealed that UA not only rescues cells from amyloid-beta-induced toxicity but also significantly enhances mitophagy, mitochondrial biogenesis and reduces oxidative stress. Furthermore, we explored the synergistic effects of a novel nutraceutical combination that includes UA, alongside other bioactive compounds known for their antioxidant and anti-inflammatory properties1. This combination was shown to amplify the mitophagic response, promoting greater cellular resilience against amyloid-beta-induced toxicity2. The results suggest that the combined intake of UA with complementary nutraceuticals could offer a comprehensive strategy for enhancing mitochondrial health and preventing the onset of AD. This research underscores the importance of dietary interventions in promoting cellular health through targeted mitophagy and offers a promising avenue for developing functional foods aimed at improving longevity and quality of life. Further in vitro and in vivo studies are underway to elucidate the underlying mechanisms and to optimize the nutraceutical combination for clinical applications against AD.
